Even small differences in plate bottom thickness can noticeably impact your imaging results. Many high-NA microscope objectives are optimized for a bottom thickness of around 170 µm - similar to a standard coverslip.
Deviate from that, and things start to change.
What happens when the bottom gets too thick?
