5×PHEM Buffer[𝗣𝗿𝗼𝗱𝘂𝗰𝘁 𝗡𝘂𝗺𝗯𝗲𝗿: 𝗣𝟯𝟭𝟳𝟱] is a pH 6.9 buffer consisting of 300 mM PIPES, 125 mM HEPES, 50 mM EGTA, and 10 mM magnesium chloride. It is filter-sterilized and can be used as PHEM buffer after 5-fold dilution with water.

When PHEM buffer was used for cytoskeletal extraction from cultured cells, cytoskeletal structures such as actin filaments and microtubules have been reported to be well preserved, allowing analysis of their three-dimensional organization and structural interactions. Recently, PHEM buffer has also been shown to be useful for sample preparation for electron microscopy.
