A recent Council for Scientific and Industrial Research (CSIR, India)-led study used RNA-seq, machine learning, and deep learning to identify biomarkers associated with progression from dengue fever to severe dengue.
We were pleased to see TRANSFAC® used in the regulatory enrichment analysis. The study reported MTF-1 and Kaiso motif signals among differentially expressed genes, linking severe dengue progression to metal ion homeostasis, oxidative stress, ferroptosis, and stress-response regulation. “Pre-vaccination immune signatures of children with inborn errors of immunity associate with COVID-19 vaccine response” published in Human Vaccines & Immunotherapeutics.
Read the full paper: vhttps://doi.org/10.1080/21645515.2026.2661495 The study addresses an important question in modern vaccinology: Why do some individuals with inborn errors of immunity respond strongly to mRNA vaccination, while others show weaker humoral responses?

By profiling immune cell subsets before vaccination, the authors showed that vaccine responsiveness is already reflected in the pre-vaccination transcriptional state of the immune system. High responders displayed stronger inducible immune programs in activated memory B cells and peripheral T follicular helper cells, including genes such as NFKB1, CD69, TIGIT, CD40L, BATF, and BTK.
We were especially pleased to see that TRANSFAC® was used and cited in the regulatory interpretation of these findings. In the study, motif analysis using TRANSFAC helped enriched transcription factor binding motifs associated with vaccine-response-related gene signatures, highlighting candidate regulators such as NF-κB1, GATA1, HNF1B, and SND1.
