Our co-founder and CEO, Scott Quainoo, saw the untapped potential of Whole Plasmid Sequencing early on, envisioning its impact beyond routine plasmid validation to transformative industrial applications.

In a pioneering 2021 publication with Enduro and Morten O. A. Sommer's group at DTU - Technical University of Denmark, Scott applied ultra-deep Whole Plasmid Sequencing to popular E. coli bioproduction strains. This revealed significant genetic heterogeneity and variants, crucial for refining biosynthetic pathways. The study urges organism engineers to avoid chassis strains hosting active insertion sequence subfamilies, such as E. coli TOP10. It also recommends monitoring for increased mutagenicity in pathway transcription initiation regions and recombinogenic repeats.

The accuracy and long-read capability of Nanopore Whole Plasmid Sequencing was key in these discoveries, revealing subtle structural variations and inversions, vital for advancing bioproduction understanding.
