Traditional metabolic research captures a snapshot after cells are lysed. But metabolic disease is dynamic, involving interconnected networks of metabolites, enzymes, and signalling molecules.

A new analysis of eighteen peer-reviewed publications reveals a significant shift: researchers are adopting bioluminescent tools that capture real-time, multi-pathway responses.

Live-cell approaches like NanoBRET and HiBiT-based assays enable researchers to watch protein behaviour and drug engagement directly in living cells. Glo-based metabolite detection assays offer faster, more sensitive detection with shared formats, making multi-pathway analysis practical.

Understanding metabolic disease requires measuring metabolism dynamically, in context, and across multiple pathways.

Do you need to detect and sort your HiBiT-tagged proteins by flow cytometry?

The Anti-HiBiT monoclonal antibody fluorophore conjugates provide sensitive, specific signal from XFD dyes.

No secondary antibody required, compatible with fixed and live-cell flow cytometry.

Available in Green 488 and Far Red 647.

Free webinar: Wednesday, 2 September 2026 at 3.00 p.m. (BST) - Hold and Kill: An Integrated Assay Strategy for RIPTAC, an Emerging Induced-Proximity Modality

RIPTACs represent a new frontier in targeted protein degradation. A heterobifunctional molecule holds a tumour-specific target and a pan-essential effector together in a ternary complex, silencing the essential protein only in cells carrying the target.

An AR-BRD4 RIPTAC is already in Phase 1/2 trials for metastatic castration-resistant prostate cancer, with PSA responses in patients who failed prior therapies.

Join Promega and ICE Bioscience to explore characterising RIPTAC mechanisms using NanoBRET® to measure ternary complex formation and assay strategies for developability assessment.

Most protein detection methods require you to kill your cells first. Fixation, lysis, blotting – just a snapshot.

HiBiT bioluminescence works differently.

The 11 amino acid HiBiT tag gives you direct, quantitative readout of your endogenous protein in live cells for protein secretion, degrader-induced protein loss, and receptor trafficking.

Our assay-ready Thaw-and-Use cells are validated for potency testing of monoclonal antibodies targeting FcγR (ADCC/ADCP), Checkpoint Inhibitors (PD-1/PD-L1, 4-1BB), and Cytokine-GF Receptors (e.g., IL-1, VEGF).

With batch sizes of up to 1,000 vials and stringent intra- and inter-batch quality controls, our Thaw-and-Use cells deliver consistent results, time after time, for QC and lot release testing.

Earlier this year at the AACR Annual Meeting 2026, Promega presented the Lumit® hKi-67 Immunoassay for cell proliferation, demonstrating how bioluminescent detection optimises high-throughput screening performance.

The assay delivers rapid, sensitive measurement of Ki-67 across suspension cells, adherent cultures, and 3D spheroid systems.

The simple, wash-free protocol with excellent sensitivity and consistency makes it ideal for compound profiling.

Missed it first time around? It's worth catching up with over the summer. The peer-reviewed abstract is now available through the AACR Journals platform.
