Our SiRNA Builder report of a potential siRNA candidates will consist from those reports:
Off Target Report
The report supports automated in silico assessment of off-target effects by comparing the siRNA sequences to the reference transcriptome. Scoring of off-target hits is based on their sequence similarity.
SNP Report
The report supports filtering out siRNAs that targeting regions with known SNP that have minor allele frequency (MAF) bigger than 0.1% of the general population.
Orthologs Report
The report will allow us to filter out sirnas that do not have full match in positions 2-18 to Target mRNAs of human and to mRNAs of potential animal model(mouse, rat, rhesus monkey and more).
miRNA Report
Filtering out sirnas that sharing same seed of known human miRNAs in order to avoid miRNAs off target.
Patent Report
Filtering out sequences with match to Patent db of all short RNA sequences (19bp - 21bp) from all the patents.
Augmented Report
the report builds all possible siRNAs with nucleotide replacements - G & C in first position in sense, A & T in last position in sense.
Algo Score Report
Prediction scoring system for siRNA efficacy that was trained on thousands of real siRNA results and was successfully validated in the past with hundreds of potent molecules. The scoring system based on several dozens of parameters such as dGs, Positions of certain letters and motifs.
