New Monkeypox Reagents Released

Monkeypox (MPX), a double-stranded DNA virus, is a zoonotic orthopoxvirus belonging to the family Poxviridae that causes disease in humans similar to smallpox with notably lower mortality. MPX is endemic to western and central Africa, with outbreaks in the Western Hemisphere associated with the exotic pet trade and international travel. The monkeypox virus or MPXV is extremely pathogenic and can spread both from animals to humans and from human to human.
As of August 9, 2022, 31,800 monkeypox cases have been reported in at least 89 territories globally -- the vast majority in places where the virus is non-endemic, according to the US Centers for Disease Control and Prevention (CDC). This is the first time that so many monkeypox cases have been reported in non-endemic and endemic countries in the world. The World Health Organization (WHO) has declared Monkeypox as Global health emergency. Abeomics has developed a line of products that would help in monkeypox research.

Ask1 (apoptosis signal-regulating kinase 1), a serine/threonine kinase, is a member of the MAPKKK family that constitutes the JNK and p38 (MAPK) cascades. Members of this family are activated by physiological and cytotoxic stresses and induce various stress responses including apoptosis. Specifically, Ask1-MAPK cascades have been shown to be involved in endoplasmic reticululum (ER) stress-induced apoptosis and in neuronal cell death. Studies suggest that ER stress-induced apoptosis plays critical roles in the pathogenesis in some neurodegenerative diseases including Alzheimer's, Parkinson's, polyglutamine (polyQ), amyotrophic lateral sclerosis (ALS) and Prion diseases. However, the regulation and function of Ask1 in neuronal cell death remains to be fully elucidated. In addition to playing a role in ER stress-induced apoptosis, Ask1 is also thought to be important in cytokine-induced apoptosis, and may also have functions other than apoptosis including roles in cell survival and differentiation. Structurally, human Ask1 has a serine/threonine kinase domain in the middle of the molecule. The kinase domain is evolutionarily conserved from nematode to human. A threonine residue with the activation loop of the kinases domain (Thr838 of human Ask1) is essential for Ask1 activation. There are two coiled-coil domains, one in the N-terminal and the other in the C-terminal domain. The coiled-coil domain in the C-terminal domain has been shown to be required for homo-oligomerzation.

ASC2/POP1 is a PAAD domain only protein originally identified in a bioinformatics screen aimed at understanding molecular apoptosis mechanisms .Human ASC2/POP1 is an 89 amino acid protein and migrates at ~10-12 kDa on SDS-PAGE gels. ASC2/POP1 has high amino acid sequence homology with ASC (64%), hence it was originally termed ASC2. The PAAD (also known as PYRIN) domain is a conserved sequence motif identified in more than 35 human proteins with putative functions in apoptosis and inflammatory signaling pathways. PAAD was named after the protein families from which it was discovered: pyrin, AIM (absent-in-melanoma), ASC [apoptosis-associated speck-like protein containing a caspase recruitment domain (CARD], and death-domain (DD)-like. In humans, the gene encoding ASC2/POP1 is on chromosome 16p12.1, only 14 kbp away from the ASC locus. The close proximity of ASC2/POP1 to ASC as well as the high sequence homology between them suggest that the ASC2/POP1 and ASC genes arose by gene duplication. Studies have shown that ASC2/POP1 associates with ASC via PADD-PADD interactions and modulates ASC-mediated roles in apoptosis and inflammation. ASC2/POP1 may also have a role in modulating other multidomain PAAD-containing proteins. However, the physiological relevance of ASC2/POP1 remains to be fully elucidated.

ASC [apoptosis-associated speck-like protein containing a CARD (caspase-recruitment domain)] is a bimodel protein containing a pyrin/paad domain (PYD/PAAD) and a caspase recruitment domain (CARD) ASC is also known as TMS1 (target of methylation-induced silencing). In general proteins, like ASC/TMS1, containing PYD/PAAD and CARD domains play key roles in regulating apoptosis and inflammation signaling pathways. Mutations in a number of PYD/PAAD- and CARD-containing proteins have been linked to inflammatory diseases and cancer. There is evidence that ASC/TMS1 has roles in both apoptosis and inflammation signaling pathways.With respect to inflammation, ASC/TMS1 interacts with the CARD of procaspase-1 and induces aggregation of a protein complex called the inflammasome, thereby regulating caspase-1 activation and secretion of IL-1b. Additionally, ASC/TMS1 has been found to be subjected to methylation-mediated silencing in a significant proportion of human breast tumors and other cancers, including melanomas, glioblastomas, non-small lung cancers, gastric and colorectal cancers. The loss of ASC/TMS1 expression in breast tumors and other cancers through methylation-mediated (epigenetic) silencing suggests that ASC/TMS1 has a role in tumorigenesis. In general, it is thought that methylation-mediated gene silencing contributes to tumorigenesis by inactivating genes involved in tumor suppression, and by conferring resistance to cell death signals by silencing genes that promote apoptosis. Thus methylation-mediated silencing of ASC/TMS1 may confer a survival advantage to tumor cells by enabling them to escape apoptosis. However, the precise role of ASC/TMS1 in the pathogenesis of cancer remains to be fully elucidated.
