If you want strong, healthy fungal cultures, starting with a sterile and nutrient-rich agar medium is the absolute key to success. Here is a quick step-by-step breakdown of the process:
1️⃣ Agar Media Preparation: Mix agar-agar powder with nutrients (like light malt extract) and water in an Erlenmeyer flask, stirring on a magnetic plate until fully dissolved.
2️⃣ Sterilization (Autoclaving): Seal the flask tightly with foil and pressure cook/autoclave it at 15 PSI (121°C) for 15–20 minutes to eliminate any contaminants.
3️⃣ Pouring in Flow Hood: Once the media cools to around 45–50°C, pour it aseptically into sterile Petri dishes inside a laminar flow hood or Still Air Box (SAB).
4️⃣ Ready for Inoculation: Let the plates cool and solidify. Once set, wrap the edges with Parafilm, and your dishes are officially ready for liquid culture, spore syringes, or tissue transfers!
