It's a clearing day! Gotto say, operating TC would always introduce dead plant in your lab. Weather it's from contamination, improper storage, lack of nutrients, or simply... forgot about them.

Especially preservative-free process (non-PPM), it's totally normal to have some plants die from being contaminated.

To use or not to use PPM? This has become a topic to debate for. Personally, I don't...(as a standard process) but only if necessarily. Coz I don't want to hide anything and have it pop up on my customer's hands....well, let's end it here for now.

What to do with the undesirable plants? I simply sterilize them before cleaning the bottles. Then resterilize them via autoclave again to make sure the bottles are free of microbs.

What are your methods? Feel free to share and don't hesitate to educate me freely.

Plant sterilization process. We were operating in laminar flow hood, ensuring clean environment for our lovely plants.

Well, truthfully, we don't really love plants, just don't wanna get so much headache in the following steps. Dealing with contamination is no fun.

Common process but EXTREMELY CRUTIAL!
I'm sharing what I do, right after I got the plants for the TC process.

First, select good mother plants. Look for the ones with good and desirable characteristics, then look out for any kind of infections or diseases. Basically, the better the mother, the better the child.

Secondly, try your best to prepare them. See if they are strong and well then clean with soap and water roughly, rid off of fungus&bac with chemicals. Then you can process to extract the explants ( the growth parts).

Well, honestly, this is not the whole thing just yet. Still more work to do but I'm happy to share a glimpse of the really serious process.

What is your packing method? My standard method would be

1st layer: packing in a plastic bag. Heat-sealed the opening and make sure there is some air inside. AND YES, it's all done inside a flow hood. Then spray them with 70%alc.

2nd layer: vacuum-sealed them for 2 purposes. Firstly, the air pocket in the inner bag would be squeeze and create an air cushion. Secondly, the inner bag may leave some gels from transferring plants into it. When I vacuum the bag, there will be no air left, reducing growth of contamination.

3rd layer: simply put all bags into a foam box for impact and temp. shield.

Anything I should add? Or I overdo it? Feel free to discuss and share your knowledge.^^
