Phage Display Libraries

Phage display is a high-throughput technology that is used to discover polypeptide-based ligands. The phage particles are genetically engineered to display random sequences on the viral coat proteins. Smith at the University of Missouri, for which he was later awarded the Nobel Prize in Chemistry (2018). Specifically, he used filamentous phage to create a library of random

displayed peptides. We currently carry a linear 15-mer peptide library (fUSE5) based on the original phage display systems by Dr. George Smith. This random peptide library exhibits excellent diversity and is proven to result in selection of high-specificity ligands with numerous research articles evidencing its efficacy. Phage display is a powerful tool in drug discovery to select peptides with high binding affinity for their target molecule. However, peptides selected against recombinant proteins often have poor pharmacokinetics, which quickly turns a promising lead into a failure.

To overcome this, at Cell Origins we use a multi-tier selection process where we build the desired in vivo behavior of the peptide into the phage display strategy. We work directly with each client to get an in-depth understanding of the biomarker and disease mechanism. This advanced research strategy is worthwhile since the resulting peptide leads have high binding affinity and exhibit excellent pharmacokinetics.
