Pattern 1 (PATN1) gets credit for spots, but it can’t make a single one! 😧

Pattern-1 (PATN1) is strongly association with a variant of the RFWD3 gene that modifies the expression of leopard complex spotting in several breeds (not just Appaloosas)! RFWD3 is an ubiquitin ligase that functions in the DNA damage response and DNA replication control. The PATN1 associated variant is located within the 3' untranslated region and thus may regulate the expression of RFWD3. Fun Fact: There are no known mutations in other species!

On its own, PATN1 does nothing visually, a horse can carry PATN1 and show no spotting at all. But when PATN1 and LP are inherited together, the results can be striking. Take a look at the beautiful Knapstrupper mare pictured, On Safari (owned by Big Vine Ranch). Think of PATN1 as an amplifier!

Here’s a basic exampke of how the interaction works (visit EtalonDx.com for more detailed information):
• LP without PATN1
→ Often minimal white
→ Often subtle LP expression
→ Sometimes no visible spotting

• LP with PATN1
→ Leopard spotting
→ Large blankets
→ Snowcap or extensive white patterns

This interaction explains why some leopard-complex horses are lightly marked while others display bold, eye-catching patterns, even within the same bloodline. Every horse is unique!

Flashn My Assets is a Reserve Champion & Futurity Champion Bay Leopard Appaloosa stallion! In addition to one copy of Leopard (LP/n) and homozygous PATN1 (PATN1/PATN1), he also carries one copy of Splashed White 4 (SW4/n) and one copy of Dominant White 20 (W20/n)! A horse can carry the Leopard allele (LP) and never have a single spot. But, without LP, leopard spotting cannot exist.
A horse must inherit at least one copy of LP to display leopard-associated traits. But LP doesn’t always mean spots or dramatic patterning! A horse can carry LP, and appear completely solid. Two LP-carrier horses can look entirely different, which is why appearance alone doesn’t tell the full genetic story!

Some LP carriers can appear solid or show subtle indicators, such as:
• Mottled skin around the muzzle or genitalia
• White sclera visible around the eyes
• Vertically striped hooves

While the coat itself may appear completely solid, genetics may say otherwise! This is true for several modifiers and white genes.

This variability is exactly why genetic testing matters, what you see (or don’t see) isn’t always what’s there.

Did you know: Genetic health risks and conditions are NOT rare in horses! 🐴

Serious genetic disorders can silently affect both stallions and mares, as they can be carriers for several genetic risks without even being unaffected.

For example Fragile Foal Syndrome (FFS), previously known as Warmblood Fragile Foal Syndrome as it was first found in Warmbloods, is a fatal, inherited condition that causes extreme skin fragility and usually results in foals being stillborn or euthanized shortly after birth (Center for Equine Health). In Warmbloods, 9–11% of horses carry the FFS mutation (Center for Equine Health). If two carriers are bred together, there’s a 25% chance each resulting foal will be affected by this lethal condition (Center for Equine Health).

Interestingly even with the name Warmblood Fragile Foal Syndrome, FFS is found in a number of other breeds, such as Thoroughbreds, Haflingers, Rocky Mountain Horses, Quarter Horses, etc. In a population of over 7000+ stock type horses, Fragile Foal Syndrome was 2x more prevalent than Malignant Hyperthermia (MH), which is required in the Quarter Horse 7 disease panel.

Testing before breeding identifies carriers so breeders can avoid carrier-to-carrier matings, the only scenario that produces affected foals.

Based on a study in the USA, 8.4% of Arabian horses carry the Severe combined immunodeficiency disease (SCID) gene.

Severe Combined Immunodeficiency (SCID) is a genetic disease where a foal is born with little to no functional immune system. These foals lack critical immune cells (B and T lymphocytes), making them unable to fight even common infections. The disease is universally fatal in horses, usually within 4-6 months of life.
