Vaccinex

Vaccinex Vaccinex is pioneering a differentiated approach to treating cancer & neurodegenerative diseases.

Vaccinex (NASDAQ: VCNX) is a clinical stage biotechnology company pioneering a differentiated approach to treating cancer and progressive neurodegenerative diseases through the inhibition of semaphorin 4D (SEMA4D). The Company’s lead drug candidate, pepinemab, blocks SEMA4D, a potent biological effector that it believes prevents immune infiltration into tumors and triggers damaging inflammation in

chronic diseases of the brain. Pepinemab is being evaluated in a Phase 1b/2 study in recurrent or metastatic head and neck cancer and in a Phase 1/2a study in Alzheimer’s Disease, with ongoing exploration of potential Phase 3 development in Huntington’s disease. The Company has also developed a proprietary drug discovery platform, ActivMAb®, that it is leveraging thru strategic collaborations, particularly by exploiting its unique capability to select high value antibodies against important multi-pass membrane receptors.

Research featuring Vaccinex’s anti-SEMA4D antibody pepinemab was highlighted in a top 10% most-viewed Alzheimer’s & Dementia paper of 2024, published on behalf of the . The publication highlights emerging advances in neuroimmunity, including SEMA4D signaling, glial biology, and neuroinflammatory mechanisms in Alzheimer’s disease.

The recognition reflects growing momentum behind targeting SEMA4D-driven neuroinflammation and glial dysfunction in Alzheimer’s disease.

We look forward to joining the conversation at .

Heading to ?
Vaccinex is advancing development of pepinemab, an anti-SEMA4D antibody, in an enlarged Phase 2b clinical trial for the treatment of Alzheimer’s disease.
Evidence suggests that Pepinemab:
• Blocks reactive gliosis contributing to neuroinflammation
• Preserves vascular integrity in brain
• Downregulates expression of AD-related proteins
Clinical and multiomics data support that treatment early in disease has favorable effects on biomarkers and appears to slow cognitive decline.

Vaccinex is excited to share a new study published in Cell Reports revealing that disruption of the SEMA4D immune-suppressive barrier can enhance trafficking of dendritic cell immunotherapy and promote systemic anti-tumor immunity in breast cancer models.

We are honored to contribute to this collaborative effort with an outstanding team of investigators at Moffitt Cancer Center and partner institutions.

Pepinemab offers a new way forward in Alzheimer’s treatment.
By targeting neuroinflammation, Pepinemab interrupts a chain of damaging processes—tau accumulation, vascular compromise, and synaptic loss—that accelerate disease progression.
